Calophyllum wallichianum, a Calophyllaceae family plant, is an endemic plant in Bangka Island, Indonesia. Chromanone acids, an emblematic group of the chemical marker of Calophyllum, have attracted researchers due to their intriguing chemical structures and diverse bioactivities. Data on the structure-activity relationship of chromanone acids against cervical cancer (HeLa) and anti-plasmodial is limited. The study explores the potential of chromanone acids from C. wallichianum stem bark against HeLa cells and anti-plasmodial activity to Plasmodium falciparum strain 3D7. Two chromanone acid derivatives, caloteysmannic and calolongic acids, were isolated from C. wallichianum. The structures of chromanone acid were focused on using HRESIMS data, as well as 1D and 2D NMR spectra. Caloteysmannic acid exhibited an IC50 value of 1.96 µg/mL, which was high activity on HeLa cells and moderate activity with an IC50 value of 2.16 µg/mL on P. falciparum strain 3D7, respectively. Calolongic acid was inactive against HeLa cells and P. falciparum.
